Abstract Mucosal-associated invariant T (MAIT) cells harbor conserved T cell receptors (TCRs) recognizing riboflavin metabolites, yet exhibit substantial diversity similar to conventional memory T cells. However, the mechanisms shaping this diversity related to their thymic ontogeny remain unclear. Here we analyze 37 samples of human thymic MAIT cells across ages and compare them with other unconventional T cells, such as iNKT and γδ T cells. We find that CD27 and CD161 serve as common markers distinguishing the maturation stages of unconventional T cells such as MAIT, iNKT and Vγ9+Vδ2+ γδ T cells. Notably, CD161+ mature MAIT cells clonally expand proportionally to aging with the upregulation of genes associated with tissue residency. MAIT cell diversity is initially determined by diverse CDR3β sequences, which become reduced upon maturation. Furthermore, 25% of MAIT cells express polyclonal dual TCRα transcripts, suggesting they arise from double-positive thymocytes with random TCRα rearrangement. Collectively, these findings show that thymic MAIT cells undergo dynamic regulation of repertoire selection, similar to conventional T cells.

When working with a few molecules, the existing protein sequencing technologies lack the single-site amino acid (AA) precision required to detect abnormalities that escape genome analysis. We offer a new method for identifying AAs and short polypeptides using the signal from the surface potential and capacitance obtained from Field Effect Transistor (FET) sensors. Using a combination of the Site-binding and Gouy-Chapman-Stern (GCS) models, we found the signatures of every single AA and polypeptides. These fingerprints are based on orthogonal features such as the proton dissociation constants of each AA’s charging sites, the dielectric constant, and the effective length.

The aim of this paper is to investigate the events connected with the establishment of the Reale Galleria di Torino as a public museum in 1832 in relation to the training path of its first director, Roberto d'Azeglio, who was in voluntary exile in Paris, where he came into contact with artists from the entourage of David's pupils in the 1920s. The function of the museum for d'Azeglio is the same as that identified by Quatrémere de Quincy: an institution of high civic value strongly linked to the present. Contemporary art and ancient art can thus establish a close dialogue to consolidate the ethical and civic involvement of visitors.

OBJECTIVE This study aims to examine age differences in the relationship between frailty and depression among older adults METHODS: A total of 1789 community-dwelling older adults were recruited from eastern China. Physical frailty and depressive symptoms were assessed using the Frailty Phenotype and the 5-item Geriatric Depression Scale, respectively. RESULTS The hierarchical multiple linear regression analysis revealed that frailty was significantly related to depressive symptoms (β = 0.272, P < 0.001) and there was a significant interaction between age and frailty (β = -0.703, P < 0.001). The Johnson-Neyman plot revealed that the relationship between frailty and depressive symptoms became weaker as people aged. CONCLUSIONS Frailty is more likely to cause depressive symptoms among the young-old than among the old-old, reflecting the age-related positivity effect. This highlights that interventions on emotional regulation should particularly target the young-old to reduce the effect of frailty on depression.

Książka Marcina Marona Romantyzm i kino. Idee i wyobrażenia romantyczne w filmach polskich reżyserów z lat 1947-1990 (2019) prezentuje interpretacje jedenastu filmów, których fabuła dotyczy wydarzeń rozgrywających się między rokiem 1789 a 1849; niektóre z nich są ekranizacjami literatury romantycznej . Wyznacznikami romantyzmu okazują się tu poglądy filozoficzne, głównie kręgu jenajskiego, oraz zjawiska historyczne kształtujące ich przyswojenie i aktywizację. Filmy zostały omówione na tle przedstawionej w ostatniej części pracy recepcji romantyzmu w czasach PRL. Książka Marona, nieco statyczna w ścisłym przestrzeganiu ograniczeń chronologicznych i kompozycyjnych, dynamizuje się jednak w lekturze, prowokując do pytań wykraczających zarówno poza ramy czasowe przyjęte przez autora, jak i wyznaczony tu krąg problemów.

Abstract Background Zika virus (ZIKV) infection is a global health problem, and its complications, including congenital Zika syndrome and Guillain-Barré syndrome, constitute a continued threat to humans. Unfortunately, effective therapeutics against ZIKV infection are not available thus far. Methods We screened the compounds collection consisting of 1789 FDA-approved drugs by a computational docking method to obtain anti-ZIKV candidate compounds targeting ZIKV RNA-dependent RNA polymerase (RdRp). SPR (BIAcore) assay was employed to demonstrate the candidate compounds’ direct binding to ZIKV RdRp, and polymerase activity assay was used to determine the inhibitory effect on ZIKV RdRp-catalyzed RNA synthesis. The antiviral effects on ZIKV in vitro and in vivo were detected in infected cultured cells and in Ifnar1 −/− mice infected by ZIKV virus using plaque assay, western blotting, tissue immunofluorescence, and immunohistochemistry. Results Here, we report that a first-in-class macrocyclic antibiotic, which has been clinically used to treat Clostridium difficile infection, fidaxomicin, potently inhibits ZIKV replication in vitro and in vivo. Our data showed that fidaxomicin was effective against African and Asian lineage ZIKV in a wide variety of cell lines of various tissue origins, and prominently suppressed ZIKV infection and significantly improved survival of infected mice. In addition, fidaxomicin treatment reduced the virus load in the brains and testes, and alleviated ZIKV-associated pathological damages, such as paralysis, hunching, and neuronal necrosis in the cerebra. Furthermore, our mechanistic study showed that fidaxomicin directly bound ZIKV NS5 protein and inhibited the RNA synthesis-catalyzing activity of ZIKV RdRp. Conclusions Our data suggest that fidaxomicin may represent an effective anti-ZIKV agent. In the light that fidaxomicin is already a clinically used drug, there might be a promising prospect in the development of fidaxomicin to be an antiviral therapeutic.

O presente estudo tem por objetivo compreender, a partir do pensamento de Francisco José de Oliveira Viana, um dos principais intelectuais conservadores brasileiros da geração dos anos 1920-40, os fundamentos e o sentido do que então eles diziam ser a ‘verdadeira’ nação brasileira, ao mesmo tempo enfatizando suas origens e identidade lusas e o seu futuro corporativo como ideal de organização e desenvolvimento da sociedade brasileira. Para tanto, o texto a seguir foi dividido em três momentos: breve reflexão sobre a História como mestra da política, como a definia Oliveira Viana; discussão sobre as origens lusas da originalidade e dos males do Brasil; breves apontamentos sobre o corporativismo como futuro da nação.
