A framework for integrating neural development and social networks in adolescence

Adolescence is a developmental period characterized by increasingly complex and influential peer contexts. Concurrently, developmental changes in neural circuits, particularly those related to social cognition, affective salience, and cognitive control, contribute to individuals’ social interactions and behaviors. However, while adolescents’ behaviors and overall outcomes are influenced by the entirety of their social environments, insights from developmental and social neuroscience often come from studies of individual relationships or specific social actors. By capturing information about both adolescents’ individual relations and their larger social contexts, social network analysis offers a powerful opportunity to enhance our understanding of how social factors interact with adolescent development. In this review, we highlight the relevant features of adolescent social and neural development that should be considered when integrating social network analysis and neuroimaging methods. We focus on broad themes of adolescent development, including identity formation, peer sensitivity, and the pursuit of social goals, that serve as potential mechanisms for the relations between neural processes and social network features. With these factors in mind, we review the current research and propose future applications of these methods and theories.

DYNC1H1 variant associated with epilepsy: Expanding the phenotypic spectrum

DYNC1H1 variants are associated with peripheral neuronal dysfunction and brain morphology abnormalities resulting in neurodevelopmental delay. However, few studies have focused on the association between DYNC1H1 variants and epilepsy. Herein, we report a case of drug-resistant focal epilepsy associated with a pathogenic variant of DYNC1H1. We further summarized the clinical, genetic, and neuroimaging characteristics of patients with DYNC1H1 variant–associated epilepsy from the relevant literature. This report expands the phenotypic spectrum of DYNC1H1-related disorder to include early-onset epilepsy, which is frequently associated with neurodevelopmental delay and intellectual disability, malformations of cortical development, and neuromuscular, ophthalmic, and orthopedic involvement.

MRI factors associated with cognitive functioning after acute onset brain injury: Systematic review and meta-analysis

Impairments of memory, attention, and executive functioning are frequently reported after acute onset brain injury. MRI markers hold potential to contribute to identification of patients at risk for cognitive impairments and clarification of mechanisms. The aim of this systematic review was to summarize and value the evidence on MRI markers of memory, attention, and executive functioning after acute onset brain injury. We included ninety-eight studies, on six classes of MRI factors (location and severity of damage (n = 15), volume/atrophy (n = 36), signs of small vessel disease (n = 15), diffusion-weighted imaging measures (n = 36), resting-state functional MRI measures (n = 13), and arterial spin labeling measures (n = 1)). Three measures showed consistent results regarding their association with cognition. Smaller hippocampal volume was associated with worse memory in fourteen studies (pooled correlation 0.58 [95% CI: 0.46–0.68] for whole, 0.11 [95% CI: 0.04–0.19] for left, and 0.34 [95% CI: 0.17–0.49] for right hippocampus). Lower fractional anisotropy in cingulum and fornix was associated with worse memory in six and five studies (pooled correlation 0.20 [95% CI: 0.08–0.32] and 0.29 [95% CI: 0.20–0.37], respectively). Lower functional connectivity within the default-mode network was associated with worse cognition in four studies. In conclusion, hippocampal volume, fractional anisotropy in cingulum and fornix, and functional connectivity within the default-mode network showed consistent associations with cognitive performance in all types of acute onset brain injury. External validation and cut off values for predicting cognitive impairments are needed for clinical implementation.

Does pubertal stage mediate the association between family environment and structure and function of the amygdala-mPFC circuit? A replication study of the longitudinal ABCD cohort

Psychosocial acceleration theory suggests that early stress accelerates pubertal development. Using half of the baseline Adolescent Brain and Cognitive Development (ABCD) cohort, Thijssen et al. (2020) provide support that accelerated puberty following stressful family environments may promote neurodevelopment. Here, we replicate and extend those analyses using 1) data from the second half of the ABCD sample (n = 3300 +, ages 9–10), and 2) longitudinal imaging data from the original sample (n = 1800 +, ages 11–12). A family environment latent variable was created and related to anterior cingulate cortex (ACC) thickness, area, white matter fractional anisotropy, amygdala volume, and cingulo-opercular network (CON)–amygdala resting-state functional connectivity. Results from the independent sample replicate the mediating effects of family environment through pubertal stage on amygdala-CON functional connectivity. Sex-stratified analyses show indirect effects via pubertal stage in girls; boys show evidence for direct associations. Analyses using wave 2 imaging data or wave 2-wave 1 difference scores from the originally-analyzed sample replicate the resting-state indirect effects. The current paper replicates the mediating role for puberty in the association between family environment and neurodevelopment. As both direct and indirect associations were found, puberty may be one of multiple mechanisms driving accelerated neurodevelopment following environmental stress.

Imaging the rapidly developing brain: Current challenges for MRI studies in the first five years of life

Rapid and widespread changes in brain anatomy and physiology in the first five years of life present substantial challenges for developmental structural, functional, and diffusion MRI studies. One persistent challenge is that methods best suited to earlier developmental stages are suboptimal for later stages, which engenders a trade-off between using different, but age-appropriate, methods for different developmental stages or identical methods across stages. Both options have potential benefits, but also biases, as pipelines for each developmental stage can be matched on methods or the age-appropriateness of methods, but not both. This review describes the data acquisition, processing, and analysis challenges that introduce these potential biases and attempts to elucidate decisions and make recommendations that would optimize developmental comparisons.

The Role of the Right Hemisphere in Processing Phonetic Variability Between Talkers

AbstractNeurobiological models of speech perception posit that both left and right posterior temporal brain regions are involved in the early auditory analysis of speech sounds. However, frank deficits in speech perception are not readily observed in individuals with right hemisphere damage. Instead, damage to the right hemisphere is often associated with impairments in vocal identity processing. Herein lies an apparent paradox: The mapping between acoustics and speech sound categories can vary substantially across talkers, so why might right hemisphere damage selectively impair vocal identity processing without obvious effects on speech perception? In this review, I attempt to clarify the role of the right hemisphere in speech perception through a careful consideration of its role in processing vocal identity. I review evidence showing that right posterior superior temporal, right anterior superior temporal, and right inferior / middle frontal regions all play distinct roles in vocal identity processing. In considering the implications of these findings for neurobiological accounts of speech perception, I argue that the recruitment of right posterior superior temporal cortex during speech perception may specifically reflect the process of conditioning phonetic identity on talker information. I suggest that the relative lack of involvement of other right hemisphere regions in speech perception may be because speech perception does not necessarily place a high burden on talker processing systems, and I argue that the extant literature hints at potential subclinical impairments in the speech perception abilities of individuals with right hemisphere damage.

The Concept of “Non-Subjectivity” in Science, Culture and Philosophy: Searching for New Meanings

The article reveals the content of some theories based on the methodological and worldview basis of the concept of “non-subjectivity”: postmodernism, constructivism, physicalism, cognitivism, neurophysiology, neuropsychology, neurolinguistics and other sciences. The heuristic aspects of these theories in the understanding of man and consciousness are shown, as well as significant shortcomings, which consist in the violation of the dialectic of objective and subjective reality, subject and object, are reveled. There are some examples, that illustrate the need for a comprehensive study of people not only on the basis of the study of the psychology of higher nervous activity, but also the activity and communication mechanisms of human study in culture and society.

Lacticaseibacillus paracasei Lpc-37® improves psychological and physiological markers of stress and anxiety in healthy adults: a randomized, double-blind, placebo-controlled and parallel clinical trial (the Sisu study)

Chronic stress is a risk-factor for the development of mood and stress-related disorders. Clinical evidence indicates that probiotics can influence the stress response and mood. The Sisu study investigated whether Lacticaseibacillus paracasei Lpc-37® (Lpc-37®) could modulate stress, mood and well-being. Prior to a two-week run-in period, 120 healthy adults (18-45 y) were stratified for sex and chronic stress and randomized to either 1.75 × 1010 colony forming units (CFU) of Lpc-37 or placebo (1:1) per day for 5 weeks. The primary objective was the effect of Lpc-37 on heart rate (HR) in response to the Trier Social Stress Test (TSST). Secondary objectives were assessed by biomarkers and self-report scales over the study. The primary hypothesis was not met in either the Intention-to-Treat (ITT) or Per Protocol (PP) population, but Lpc-37 reduced the increase in HR in participants with low chronic stress (LCS) and increased HR in participants with high chronic stress (HCS) during the TSST. Supporting significant efficacy in the PP population (n = 113), Lpc-37 reduced perceived stress following intervention. More significant effects were identified within the subgroups where Lpc-37 reduced exhaustion during the TSST and normalized cortisol levels at 8pm in participants with LCS, reduced perceived stress also in females, and increased perceived health and sleep-related recovery in participants with HCS. Adverse events (AEs) were similar between groups, there were no severe AEs, and vital signs remained unchanged. Overall, Lpc-37 reduced perceived stress compared to placebo. Other beneficial effects within biomarkers related to stress indicate that the effects of Lpc-37 may be differentially dependent on sex and chronic stress. (ClinicalTrials.gov: NCT03494725).

Behavioral tests in rodent models of stroke

Rodents are the most widely used experimental animals in stroke research due to their similar vascular anatomy, high reproductive rates, and availability of transgenic models. However, the difficulties in assessing higher brain functions, such as cognition and memory, in rodents decrease the translational potential of these studies. In this article, we review commonly used motor/sensorimotor and cognition tests in rodent models of stroke. Specifically, we first briefly introduce the objective and procedure of each behavioral test. Next, we summarize the application of each test in both ischemic stroke and hemorrhagic stroke. Last, the advantages and disadvantages of these tests in assessing stroke outcome are discussed. This review summarizes commonly used behavioral tests in stroke studies and compares their applications in different stroke types.

Dopamine response gene pathways in dorsal striatum MSNs from a gene expression viewpoint: cAMP-mediated gene networks

Abstract Background Medium spiny neurons (MSNs) comprise the main body (95% in mouse) of the dorsal striatum neurons and represent dopaminoceptive GABAergic neurons. The cAMP (cyclic Adenosine MonoPhosphate)—mediated cascade of excitation and inhibition responses observed in MSN intracellular signal transduction is crucial for neuroscience research due to its involvement in the motor and behavioral functions. In particular, all types of addictions are related to MSNs. Shedding the light on the mechanics of the above-mentioned cascade is of primary importance for this research domain. Results A mouse model of chronic social conflicts in daily agonistic interactions was used to analyze dorsal striatum neurons genes implicated in cAMP-mediated phosphorylation activation pathways specific for MSNs. Based on expression correlation analysis, we succeeded in dissecting Drd1- and Drd2-dopaminoceptive neurons (D1 and D2, correspondingly) gene pathways. We also found that D1 neurons genes clustering are split into two oppositely correlated states, passive and active ones, the latter apparently corresponding to D1 firing stage upon protein kinase A (PKA) activation. We observed that under defeat stress in chronic social conflicts the loser mice manifest overall depression of dopamine-mediated MSNs activity resulting in previously reported reduced motor activity, while the aggressive mice with positive fighting experience (aggressive mice) feature an increase in both D1-active phase and D2 MSNs genes expression leading to hyperactive behavior pattern corresponded by us before. Based on the alternative transcript isoforms expression analysis, it was assumed that many genes (Drd1, Adora1, Pde10, Ppp1r1b, Gnal), specifically those in D1 neurons, apparently remain transcriptionally repressed via the reversible mechanism of promoter CpG island silencing, resulting in alternative promoter usage following profound reduction in their expression rate. Conclusion Based on the animal stress model dorsal striatum pooled tissue RNA-Seq data restricted to cAMP related genes subset we elucidated MSNs steady states exhaustive projection for the first time. We correspond the existence of D1 active state not explicitly outlined before, and connected with dynamic dopamine neurotransmission cycles. Consequently, we were also able to indicate an oscillated postsynaptic dopamine vs glutamate action pattern in the course of the neurotransmission cycles.

Family drawing and psychological vulnerability in Children’s representations of parental divorce

Parental divorce is a traumatic experience for children, a stressful life event which may contributes to a future psychological vulnerability. The aim of this study is to explore psychological distress in children after parents’ separation. This research was based on a convenience sample of one-hundred children, divided in two subgroups: (1) 50 coming from a separated family (observational sample) with an age range spanning from 7 to 12 years [Mean age = 9.2 ± 1.5] and (2) 50 belonging to a nuclear family (control sample) with the same range of age [Mean age = 9.5 ± 1.6]. The sample resulted homogeneous for the gender: in particular, the observational group, is composed of 30 females (60%) and 20 males (40%). Showed several differences, highlighting the presence of negative affectivity, a well-known risk factor for the development of a depressive disorder, by reflecting the possible link between home environment and self-perception. In conclusion, this instrument, like any other projective proof, constitutes a privileged access to the psychological world in which the drawing remains the best communication channel and provides us precious informations about the emotional state of the children.

Neural activity reveals effects of aging on inhibitory processes during word retrieval

ABSTRACT Word retrieval difficulties are one of the most frustrating problems for older adults. Poorer access to phonological representation of the target word has been postulated as the underlying deficit, supported by findings of improved word retrieval after phonological priming. To better understand aging effects in the underlying neurophysiology associated with word retrieval, this study examined electrophysiological correlates of phonological priming and word retrieval in adults. Young, middle-aged, and older adults viewed pictures that were preceded by pseudo-word primes that either shared initial phonemes with or were unrelated to the picture’s name. Participants made phonological judgments regarding the prime and picture prior to naming the picture. Behavioral and event-related potential correlates of phonological priming and word retrieval were recorded. All groups benefitted similarly from phonological priming, evidenced by faster phonological judgment response times and increased ease of word retrieval for primed pictures, indexed by the N400 priming effect. The peak latency of the N2, however, showed an incremental delay with age. High correlation between N2 peak latency and clinical measures of inhibition suggested an age-related delay in the inhibition of primed lexical competitors. Taken together, our results indicated intact activation of phonological representation of the picture’s name but age-related delays in inhibition of primed competitors. Interestingly, our findings revealed that delays in inhibiting lexical competitors may begin as early as middle age, highlighting the importance of including multiple age groups to better represent the aging trajectory.

Habilidades previas para el cálculo y para la escritura, y vocabulario receptivo en niños de seis años con trastorno por déficit de atención e hiperactividad (TDAH).

Objetivos. Estudiar la correlación entre las habilidades previas para el cálculo y para la escritura, y el vocabulario receptivo en niños de seis años con trastorno por déficit de atención e hiperactividad (TDAH). Pacientes y métodos. 16 niños con TDAH -de 6 años de edad- , a los cuales se les administró las dimensiones de Habilidades previas para el cálculo (HPC) y Habilidades previas para la escritura (HPE) de la Evaluación de Habilidades Básicas para el Aprendizaje (EHBA) y el Test Figura-Palabra de Vocabulario Receptivo, llevándose a cabo un análisis de tipo correlacional. Resultados. Se encontró que la correlación tanto a nivel de las HPC y el vocabulario receptivo, como a nivel de las HPE y este último constructo, resultó estadísticamente positiva, aunque baja; esto sucedió debido a la existencia de otros factores afines, de naturaleza tanto extrínseca como intrínseca, presentes en esta población en particular. Conclusiones. En cuanto a la relación existente entre las HPC y el vocabulario receptivo, el resultado describió que tanto los conocimientos previos para el cálculo (Vg.: Nociones básicas de aritmética, razonamiento numérico) como el soporte léxico-semántico resultaron significativamente bajos; posiblemente debido a la existencia de algún evento encubierto y/o condición de naturaleza ambiental. Un resultado similar se encontró también a nivel de la correlación entre las HPE y el vocabulario receptivo, donde ambos eventos revelaron un compromiso tanto en relación a los conocimientos previos para el desarrollo de la escritura (Vg.: Nociones básicas de grafías y grupos silábicos), como en el acceso léxico-semántico

Pioglitazone, a PPARγ agonist rescues depression associated with obesity using chronic unpredictable mild stress model in experimental mice

Pioglitazone, a peroxisome proliferator activated receptor gamma (PPARγ) agonist belonging to thiazolidinedione class, is mainly used in diabetes mellitus. Obese subjects are twice likely to become depressed than non-obese individuals. The biological mechanisms linking depression with obesity still remain poorly understood and there is immense need for better therapeutic intervention against such co-morbid disorders. The present study investigates the effect of pioglitazone on the chronic unpredictable mild stress (CUMS) induced depression in obese mice by using behavioral tests and biochemical estimations. Mice were fed with high fat diet (HFD) for 14 weeks and were further subjected to different stress procedures for 28 days to induce depressive behavior. Animals were administered orally with pioglitazone (30 mg/kg p.o.)/escitalopram (10 mg/kg p.o.)/vehicle (10 ml/kg p.o.) daily from day 15–28. Various behavioral paradigms such as sucrose preference test, forced swim test (FST), tail suspension test (TST) and elevated plus maze (EPM) were performed. Biochemical estimations including plasma glucose, total cholesterol, triglycerides, and total proteins were performed. The data obtained from behavioral assays and biochemical assessments indicated that obese animals exhibited severe depressive-like behavior compared to non-obese animals. Furthermore, obese animals subjected to CUMS worsen the depressive behavior compared to obese control animals. Repetitive treatment with pioglitazone reversed the CUMS induced behavioral and biochemical alterations in HFD fed obese mice which atleast in part may be mediated through improving altered plasma glucose. The study suggests that pioglitazone needs further attention with respect to molecular mechanisms that could provide a better therapeutic strategy against depression associated with obesity.

The microbiome: A key regulator of stress and neuroinflammation

There is a growing emphasis on the relationship between the complexity and diversity of the microorganisms that inhabit our gut (human gastrointestinal microbiota) and health/disease, including brain health and disorders of the central nervous system. The microbiota-gut-brain axis is a dynamic matrix of tissues and organs including the brain, glands, gut, immune cells and gastrointestinal microbiota that communicate in a complex multidirectional manner to maintain homeostasis. Changes in this environment can lead to a broad spectrum of physiological and behavioural effects including hypothalamic-pituitary-adrenal (HPA) axis activation, and altered activity of neurotransmitter systems and immune function. While an appropriate, co-ordinated physiological response, such as an immune or stress response are necessary for survival, a dysfunctional response can be detrimental to the host contributing to the development of a number of CNS disorders. In this review, the involvement of the gastrointestinal microbiota in stress-mediated and immune-mediated modulation of neuroendocrine, immune and neurotransmitter systems and the consequential behaviour is considered. We also focus on the mechanisms by which commensal gut microbiota can regulate neuroinflammation and further aim to exploit our understanding of their role in stress-related disorders as a consequence of neuroinflammatory processes.

Self-harm and suicidal acts: a suitable case for treatment of impulsivity-driven behaviour with repetitive transcranial magnetic stimulation (rTMS)

Summary Suicidal thinking, self-harm and suicidal acts are common, although determining their precise prevalence is complex. Epidemiological work has identified a number of associated demographic and clinical factors, though, with the exception of past acts of self-harm, these are non-specific and weak future predictors. There is a critical need shift focus from managing ‘suicidality-by-proxy’ through general mental health treatments, to better understand the neuropsychology and neurophysiology of such behaviour to guide targeted interventions. The model of the cognitive control of emotion (MCCE) offers such a paradigm, with an underlying pan-diagnostic pathophysiology of a hypoactive prefrontal cortex failing to suitably inhibit an overactive threat-responding limbic system. The result is a phenotype – from any number of causative gene–environment interactions – primed to impulsively self-harm. We argue that such neural dysconnectivity is open to potential therapeutic modification from repetitive transcranial magnetic stimulation (rTMS). The current evidence base for this is undoubtedly extremely limited, but the societal and clinical burden self-harm and suicide pose warrants such investigation.
