TITHING DISCOURSE IN THE DYNAMICS OF THE REGIONAL ASSEMBLY OF THE PENTECOSTAL CHURCH in INDONESIA (GPdI) CENTRAL KALIMANTAN

The objectives of this study are, to analyze and reveal the forms of tithing discourse, the underlying ideologies, and their implications within the dynamics of the Regional Assembly of the Pentecostal Church of Indonesia in Central Kalimantan. Initially, tithing was spiritual in nature, but it has undergone significant transformations alongside the development of the church as an organization—shifting from an offering to God, into an offering made as obedience to the church institution, and even as a requirement for attaining certain positions within the church organization. This research employs a qualitative method with data collection techniques including observation, interviews, and document study. The theoretical frameworks applied are the theory of power–knowledge relations, hegemony theory, and genetic structuralism. The findings demonstrate that the forms of tithing discourse within the dynamics of the GPdI Regional Assembly in Central Kalimantan are the Biblical discourse can be traced from the history of tithing in the Bible, GPdI theology on tithing becomes a doctrine of belief. The ideologies behind the tithing discourse in the dynamics of the Regional Assembly of the Pentecostal Church in Indonesia Central Kalimantan are Political Ideology, Economic Ideology, Social Ideology. The implications of the tithing discourse in the dynamics of the Regional Assembly of the Pentecostal Church in Indonesia Central Kalimantan are, to The GPdI Central Kalimantan Regional Assembly 2017-2022 period, to The GPdI Central Kalimantan Regional Assembly 2022-2027 period, Amendment of the GPdI Articles of Association and Bylaws of 2012, and to the GPdI Central Kalimantan Pastors.

HINDU MARRIAGE RITUALS IN DENPASAR: COMMODIFICATION IN A GLOBALIZED WORLD

A prominent example of globalization's effect on local traditions can be seen in Denpasar's marriage rituals, where modern global culture often vies with a rich local heritage. This phenomenon has implications for transforming values ​​in society’s culture to the private realm in the marriage ceremony as a religious practice, which is now one of the economic potentials through commodification. This study focuses on the commodification of Hindu marriage in Denpasar City in the global era. This study aims to find out what forms and how it occurs. This study is a qualitative study with a cultural studies paradigm. Data is collected through observation, interview, and document study methods. The theory of Commodification and social practices are used to analyze the problems of the study. Data analysis is carried out descriptively and interpretatively by describing the phenomena that occur critically. The study results show that the forms of commodification found in marriage in the global era in Denpasar City are socio-cultural and economic. TOURISTIFICATION OF THE PURA AGUNG BESAKIH AREA AS A CULTURAL TOURISM DESTINATION

Cultural commodification to the touristification of sacred areas is now increasingly commonplace on the island of Bali. The existence of temples that were initially only used for religious ritual activities has slowly begun to experience multiple functions and become a tourist attraction in the development of tourism in Bali. In Bali, many holy places have experienced tourism, including the Pura Agung Besakih also known as The Great Temple of Besakih. This research seeks to reveal the factors that affect the tourism of the Pura Agung Besakih area, the forms of tourism of the Pura Agung Besakih area, and the implications of what happens with the tourism of the Pura Agung Besakih area. Field data collection was conducted by direct observation in the Pura Agung Besakih area, in-depth interviews with speakers, and literature review correlating with this research. The successfully collected data were analyzed and dissected using commodification theory and practice theory. The results show that global tourism and tourism presence seem to dominate the external background of tourism in the Pura Agung Besakih area, in addition to being strengthened by technology flows, social media, and support from stakeholder regulations that shelter. On the other hand, market ideology and attempts to take economic advantage of external phenomena motivate the occurrence of tourism in the area of ??Pura Agung Besakih. This ultimately implies temple commercialization, religiosity, profanation, degradation of cultural and economic values, and the risk of environmental damage. CULTURAL HYBRIDITY IN PRESERVING THE MACAPAT TRADITION

There is a phenomenon of urgency in the local cultural order, which causes the interaction between traditional values ??and cultural influences to become stronger. Globalization can accelerate the hybridization process, but it is feared that it will give rise to new challenges. The role of mass media is very large in ultimately creating popular culture, popular culture in each era determines the formation of hybridity. The theories used and relevant according to the research focus to answer research questions are Cultural Hybridation (Bhaba). This study aims to explore the hybridity of macapat culture in the form of presentation of the macapat tradition so that it becomes an art with a hybrid identity. So, the research question is formulated as follows: How is the hybridity of the macapat tradition in the special region of Yogyakarta? This research used the qualitative data gathered from primary and secondary sources, in order to critically examine the issue of cultural hybridity in Macapat communities Yogyakarta. SOCIAL PHENOMENON OF PRE WEDDING PHOTO TRENDS AT DENPASAR COMMUNITY WEDDING CEREMONY

The social phenomenon of pre-wedding photos has now begun to be widely discussed by some people who are starting to be sensitive to social changes in the area of Denpasar City. Infact, in Denpasar City at this time almost all bride and groom couples carry out pre-wedding photo sessions before their wedding ceremony takes place. The bride and groom couples even really focus their attention on the results of their pre-wedding photos. This eventually became a conversation that raised pros and cons among parents whose children were going to have a wedding ceremony. Some parents feel that pre-wedding photos cost too much money and costs that they should be able to spend to bail out funds when preparing a series of wedding ceremonies. In contrast to people who feel capable in terms of costs, they will certainly take the pre-wedding costs lightly compared to the satisfaction they will get later after seeing the results of the pre-wedding photos they take. The bride and groom did a pre-wedding photo session with the theme and concept they specified. When going to do a pre-wedding photo session, the bride and groom have to prepare a lot of money. Depending on the theme, concept and place they will use later. In Bali, especially in Denpasar City, the cost for the “pawiwahan”or wedding ceremony is quite high, with the addition of a new tradition, namely pre-wedding photos, it will make people spend even more. The meaning of this pre-wedding photo session is still not much can explain, prospective brides who do pre-wedding photo sessions only say they take pre-wedding photos to follow today's trends. The new trend that is entering Bali is slowly making it a mandatory tradition.

CAUSE FACTORS SYMBOLIC RESISTANCE OF GRAPHIC DESIGNERS IN DENPASAR CITY

The discourse of "free design" that was introduced by printing companies as a form of strategy in dealing with business competition, has caused controversy in the graphic design field. Indirectly, this discourse is considered to be able to influence the mindset of the community and become a negative stigma against the graphic designer profession, and the discourse is considered to be a threat to the continuity of the graphic designer profession.This study aims to answer the factors that cause resistance. The study data were taken from observation, social media, and interviews, and analyzed using Michel Foucault's power relation theory and Norman Fairclough's theory of critical discourse analysis.

Abstract 1120: α-Tocopherol protects cells from lipid peroxidation and rescues tumorigenic phenotypes in CS/CSL patients with germline SDHx variants

Abstract Cowden syndrome (CS), a Mendelian autosomal-dominant disorder, predisposes to breast, thyroid, and other cancers. Germline mutations in phosphatase and tensin homolog (PTEN) have been found in 25% of classic CS individuals accrued from the community. Additionally, we have shown that germline variations in succinate dehydrogenase genes (SDHx) occur in 8% of 608 PTEN mutation-negative CS and CS-like (CSL) individuals (SDHvar+), compared to 0/700 controls (p<0.0001, Fisher's 2-tailed exact test). SDHx genes encode for mitochondrial complex II which lies at the crossroads of electron transport and the tricarboxylic acid cycle. We previously showed that SDHx variants result in elevated reactive oxygen species (ROS) and in disruption of such mitochondrial metabolites as flavin adenine dinucleotide (FAD) and nicotinamide adenine dinucleotide (NAD) equilibrium in CS/CSL patient-derived lymphoblastoid cells. Consequently, improper NADH quinone oxidoreductase 1 (NQO1)-p53 interaction results in destabilization of p53 and apoptosis resistance. Here we further demonstrate that elevated ROS production results in higher lipid peroxidation in SDHvar+ cells compared to healthy control cells. This increased oxidative stress led to increased mutation accumulation in mitochondrial hypervariable region II (HVRII) in samples with SDHx variants compared to controls. Interestingly, α-tocopherol treatment, but not other antioxidants, can rescue SDHvar+ cells from apoptosis resistance. This lipid soluble vitamin E, functions as a peroxyl-radical scavenger, protecting SDHvar+ cells from oxidative damage. We observed down regulated lipid peroxidation in SDHvar+ cells after α-tocopherol exposure, as well as recovered p53 expression and FAD/NAD levels. We conclude that α-tocopherol selectively protects SDHvar+ cells from redox stress and downstream tumorigenic reprogramming, which may have intriguing clinical implications such as using vitamin E as a potential preventive adjunct for CS/CSL individuals with SDHx variants.
