Chemical investigations of marine sponges, Axinyssa sp., Petrosia sp. and Cacospongia mycofijiensis

Two new nitrogenous germacrane sesquiterpenes, (1Z,4Z)-7alphaH-11-aminogermacra-1(10),4-diene, and N,N'-11-bis[(1Z,4Z)-7alphaH-germacra-1(10),4-dienyl]urea were isolated from the sponge Axinyssa sp., together with a known steroid axinysterol in 0.06, 0.12, and 0.03% w/w based on the MeOH extract, respectively. The compound (1Z,4Z)-7aH-11-aminogermacra-1(10),4-diene showed significant antimicrobial activity against Staphylococcus aureus, Bacillus subtilis, and Candida albicans with inhibition zones of 23, 22, and 27 mm, respectively, at concentration of 500 microgram/disc. The chemical study of the sponge Petrosia sp. led to the isolation of two pyridoacridine alkaloids, including the new 2-bromoamphimedine and the known petrosamine, together with a new isoguanine derivative, namely 1,3,9-trimethyl-8-hydroxyisoguanine in 0.03, 1.5, and 0.01% w/w based on the n-butanol extract, respectively. Petrosamine exhibited antimicrobial activity against Bacillussubtilis with an inhibition zone of 22 mm at concentration of 200 microgram/disc and possessed acetylcholinesterase inhibitory activity with an IC[subscript 50] of 91 nM, which was six times more potent than the reference compound galanthamine. Molecular docking study indicated the quaternary ammonium group of petrosamine donated the major and strong interactions to the key amino acid residues in the active site of Torpedo californica acetylcholinesterase. Bioassay-guided fractionation led to the isolation of the actin polymerization inhibitor latrunculin A and the microtubule-stabilizing agent laulimalide from the sponge, Cacospongia mycofijiensis. The yields of latrunculin A and laulimalide were 0.54 and 0.02% w/w based on the MeOH extract, respectively. In addition, three C1-C14 fragment analogs of laulimalide, LAU13, LAU14, and LAU16, were successfully synthesized in good yields.

Verification of tidal trends in Cananeia (SP) and Santos (SP)

Recent studies indicate an imminent global rise in mean sea level. However, comprehending variations in tidal patterns is imperative before analyzing the scenario of extreme events, typically associated with the combination of high tides and intense meteorological influences. This study presents local trends in sea level, amplitude, and phase of the two main tidal components in Cananeia (SP) and Santos (SP), utilizing tide gauge data from records of sufficient length to avoid the need for nodal correction in the tide components. Using more up-to-date tide gauge data allowed the investigation of observed trends. Long-term tidal data series revealed an upward trend in mean level of approximately 4,324 mm/year in Cananeia and 0,538 mm/year in Santos. Both locations were affected by human activities along their estuaries, with Santos experiencing significant variations in the amplitude and phase values of the tidal constituents annually.

RELAÇÃO ENTRE A OBESIDADE, INATIVIDADE FÍSICA E PRESSÃO ARTERIAL EM EDUCANDOS NAS CIDADES DE BAURU, IBITINGA, JAÚ E SÃO SEBASTIÃO- SP

Arterial hypertension (AH) is a disease that affects children and adolescents and its identification, helps to prevent the premature development of cardiovascular diseases. The objective of this study was to verify the presence of blood pressure (BP) above normal and to relate it to obesity and lifestyle of scholars in the cities of Bauru, Ibitinga, Jaú and São Sebastião. A total of 369 children and adolescents were evaluated. Information on health and physical exercise were obtained through anamnesis. After 5 minutes of rest, hemodynamic and anthropometric assessments were performed on each participant. Odds rate was used to assess the risk of above-normal blood pressure in overweight and inactive children. Pearson’s correlation was used to correlate anthropometric and hemodynamic variables. Among those evaluated, 28% had altered BP values and 29% were already overweight and obese. In addition, 89% of students were classified as insufficiently active. Among overweight scholars, 48% had altered BP and, among obese, BP was altered in 65%. Overweight children and adolescents had 3.59 times more risk to have altered BP. It can be concluded that the results are extremely relevant to reinforce the inclusion of different strategies in schools for the control of obesity and sedentary lifestyle, in order to avoid early cases of hypertension and cardiovascular diseases.

PENGARUH SURIMI DARI IKAN SWANGGI (Priacanthus sp.), IKAN KURISI (Nemipterus sp.), DAN IKAN KUNIRAN (Upeneus sp.) TERHADAP KARAKTERISTIK CUMI-CUMI ANALOG

Cumi-cumi analog merupakan salah satu produk olahan dari surimi yang diciptakan untuk meniru cumi-cumi yang memiliki tekstur dan rasa yang khas. Penggunaan jenis ikan laut sebagai bahan baku surimi perlu diperhatikan untuk memperoleh cumi-cumi analog yang diminati masyarakat. Tujuan penelitian ini adalah mengetahui karakteristik surimi dan cumi-cumi analog dari jenis ikan laut yang berbeda serta mengetahui jenis ikan laut terbaik yang digunakan terhadap karakteristik cumi-cumi analog. Metode penelitian yang digunakan yaitu experimental laboratories menggunakan Rancangan Acak Lengkap (RAL) dengan perlakuan jenis ikan laut yang berbeda yaitu (A) ikan swanggi, (B) ikan kurisi, dan (C) ikan kuniran dengan tiga kali ulangan. Data parametrik dianalisis menggunakan ANOVA dan uji lanjut Tukey HSD. Data nonparametrik dianalisis dengan Kruskal-Wallis dan uji lanjut Mann-Whitney. Hasil penelitian menunjukkan bahwa jenis ikan laut terbaik berdasarkan karakteristik fisik dan kimia cumi-cumi analog diperoleh dari ikan swanggi dengan nilai gel strength surimi dan pH surimi 2.591,13±45,54 g/cm2 dan 7,12±0,08 serta cumi-cumi analog dengan nilai cohesiveness 0,44±0,01, elasticity 8,81±0,17 mm, derajat putih 77,89±0,87%, Water Holding Capacity 53,33±2,31%, kadar air 67,16±0,75%, kadar abu 15,00±0,95%, kadar protein 57,90±1,22%, kadar lemak 1,48±0,51%, pH 6,92±0,01. Jenis ikan laut terbaik berdasarkan uji hedonik diperoleh dari ikan kurisi dengan nilai selang kepercayaan tertinggi sebesar 4,46 ˂ µ < 4,70 pada tingkat kepercayaan 95%.

Pedobacter duraquae sp. nov., Pedobacter westerhofensis sp. nov., Pedobacter metabolipauper sp. nov., Pedobacter hartonius sp. nov. and Pedobacter steynii sp. nov., isolated from a hard-water rivulet

Five isolates that were related phylogenetically to members of the genus Pedobacter were isolated from freshwater of the hard-water creek Westerhöfer Bach, North Germany. The five strains (WB 2.1-25T, WB 2.3-71T, WB 3.3-3T, WB 3.3-22T and WB 2.3-45T) were Gram-negative and chemoheterotrophic, with rod-shaped cells. Most of their metabolic properties matched those given in the description of the genus Pedobacter. Consistent with the genus description, their fatty acids included mainly iso-C15 : 0 and summed feature 3 (C16 : 1 ω7c, iso-C15 : 0 2-OH or both); C16 : 1 ω5c, C16 : 0, iso-C15 : 0 3-OH, C16 : 0 3-OH and iso-C17 : 0 3-OH were present in smaller amounts. The major isoprenoid quinone was menaquinone 7. With one exception, binary similarity values of the almost complete 16S rRNA gene sequences determined among the isolates as well as between the isolates and type strains of Pedobacter species were lower than 98.5 %. The only exception was the close relationship between Pedobacter caeni DSM 16990T and strain WB 2.3-45T (99.2 % similarity). DNA–DNA reassociation values determined for this pair of strains was 29.8 %, indicating that strain WB 2.3-45T represents a unique genospecies. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strains WB 2.1-25T and WB 2.3-71T form a group that is moderately related to P. caeni and strain WB 2.3-45T (98.5 % similarity). Strains WB 3.3-3T and WB 3.3-22T (98.5 % similarity) branched separately from these four organisms. The five phylogenetically isolated strains differed from each other as well as from the type strain of the type species (Pedobacter heparinus DSM 2366T) and some related representatives of the genus in several metabolic reactions and cultural parameters. On the basis of phenotypic and phylogenetic distinctiveness, five novel species are proposed: Pedobacter duraquae sp. nov., with WB 2.1-25T (=DSM 19034T=CIP 109481T) as the type strain; Pedobacter westerhofensis sp. nov., with WB 3.3-22T (=DSM 19036T=CIP 109479T) as the type strain; Pedobacter metabolipauper sp. nov., with WB 2.3-71T (=DSM 19035T=CIP 109480T) as the type strain; Pedobacter hartonius sp. nov., with WB 3.3-3T (=DSM 19033T=CIP 109468T) as the type strain; and Pedobacter steynii sp. nov., with WB 2.3-45T (=DSM 19110T=CIP 109507T) as the type strain.
