The Machiavellian Moment

"The Machiavellian Moment" is a classic study of the consequences for modern historical and social consciousness of the ideal of the classical republic revived by Machiavelli and other thinkers of Renaissance Italy. J.G.A. Pocock suggests that Machiavelli's prime emphasis was on the moment in which the republic confronts the problem of its own instability in time, and which he calls the "Machiavellian moment."After examining this problem in the thought of Machiavelli, Guicciardini, and Giannotti, Pocock turns to the revival of republican thought in Puritan England and in Revolutionary and Federalist America. He argues that the American Revolution can be considered the last great act of civic humanism of the Renaissance. He relates the origins of modern historicism to the clash between civic, Christian, and commercial values in the thought of the eighteenth century.

Sustainable design concept for a trendy beverage container

This research presents an innovative 330-ml beverage packaging solution that merges accessibility, design, and ergonomics for user comfort. The design incorporates shape principles, clear typography, and minimalist colours and symbols, balancing visual appeal and functionality. As demand for attractive options grows, this study investigates consumer preferences through surveys and interviews. It identifies key factors influencing choices, enhancing the design through quality function deployment (QFD) and product design development (PDD) methodologies. This approach integrates shape design, typography, and colour theory to meet aesthetic and functional needs. The packaging features ‘a bear-paw-shaped watermark’ for stability and ergonomic design for comfortable handling. A minimalist label, with fruit-inspired illustrations, modern fonts, and finishes like matte or glossy, enhances visual appeal. The ‘Trendy-Bottle Design Platform’ aids creation with templates, 3D visualisation, and collaboration tools. Key elements like brand details, nutritional facts, allergen information, a recycle sign, and sustainability symbols ensure functionality and alignment with consumer needs. From a customer perspective, trendy bottles should offer ergonomic designs, spill-proof functionality, eco-friendly materials, and affordability. Manufacturing considerations include optimising shelf space, with rectangular bottles saving space despite higher costs, and rounded bottles saving material but increasing shelf costs. Collaboration ensures moulds meet aesthetic and functional needs, improving appeal.

Whey Protein Isolate and β-Lactoglobulin-Modified Alginate Hydrogel Scaffolds Enhance Cell Proliferation for Cultivated Meat Applications

Innovative changes to our current food system are needed, and one solution is cultivated meat, which uses modern engineering, materials science, and biotechnology to produce animal protein. This article highlights the advantages of incorporating whey protein isolate (WPI) and β-lactoglobulin (β-LG) into hydrogel networks to aid cell growth on cultivated meat scaffolds. The protein and polysaccharide (i.e., alginate) components of the scaffolds are food-grade and generally regarded as safe ingredients, enabling the transition to more food-safe, edible, and nutritious scaffolds. The impact of WPI and varying properties on cell performance was evaluated; alginate concentration and the addition of proteins into the hydrogels significantly altered their stiffness and strength. The results of this study demonstrate the innocuous nature of novel scaffolds and reveal enhanced cell proliferation on WPI and β-LG-modified groups compared to standard biomaterial controls. This work serves as a stepping stone for more comprehensive analyses of WPI, β-LG, and alginate scaffolds for use in cultivated meat research and production.

Automation of inspection of rolling stock at a maintenance point using a robotic complex

The article discusses the need to improve the efficiency of freight cars operation by introducing modern technologies in the maintenance process. To improve the quality of technical inspection of rolling stock at the maintenance point, it is offered to introduce a robotic complex capable of autonomously conducting technical inspection and diagnostics of rolling stock. Structurally, the robotic complex includes: chassis; manipulator; automation, control and data transmission system; diagnostic equipment; power unit. The possibility of using various diagnostic tools, vision systems, non-contact scanning sensors is considered. The functions of the complex are to pass through the inspection positions, identify malfunctions, accumulate information about the technical condition of the car with the transfer of information to the fleet operator or maintenance point.

Gender-Role Stereotypes in the Bulgarian Family: Cross-Generational Transmission of Gender Attitudes

This article reports a representative quantitative and qualitative study covering the entire territory of Bulgaria, related to the analysis of stereotypes regarding male and female roles in the family and in the society. The results of the study indicate that the factor “age” is significant in relation to attitudes towards the social roles of men and women in the modern family, including in relation to domestic violence. The interpretation of the results enabled the formulation of justified conclusions related to changes in attitudes towards the roles of men and women in a dynamic way. The hypothesis was proven that the tensions between them can be more easily overcome through appropriate differentiated measures tailored to the different age groups.

Conversion

Conversion – the turning from one religion, belief, or way of life to another informed by the saving activity of Christ – is a central theme of Christianity. From a theological perspective, conversion, the human experience of salvation, is distinguished from regeneration, the hidden, divine work of God. Although conversion is often associated with a particular strand of modern Christianity (evangelical) and a particular kind of experience (a sudden, shattering, complete event), it is, when examined over two millennia, a phenomenon far more complex than any one-dimensional profile would suggest. This entry examines Christian conversion from the perspective of the biblical record, scholarly approaches toward the phenomenon, historical episodes of conversion, and recurring themes and issues in the history of conversion.

Two Views on One Problem: The Eurasian Project and “One Belt — One Road”

The article compares the theoretical foundations of ethno-cultural policy in the Eurasian region.Aim. The goal set by the authors is to compare two approaches in the theory of social dynamics, on the basis of which the ethno-cultural policy of the modern states of the region — Russia (USSR) and China — is based.Tasks. Identification of fundamental differences in the geopolitical strategies developed by the theorists of Eurasianism and Chinese scientists.Methods. In the context of the implementation of this task — a comparison of the methodological foundations of real political projects in the Eurasian space, through logical analysis, differences are established in the theories of geopolitical dynamics proposed in the Eurasian concept and in Chinese science. The method of comparative analysis of ethno-cultural and ethno-economic policies in the Eurasian region of the two most influential powers allows us to conclude that alternative theoretical concepts and their paradigmatic limitations are adequate to real geopolitical processes.Results. The study showed that the concept of Eurasian geocivilization, which was formed as overcoming the limitations of the Slavophile version of Russian civilization in the context of the collapse of traditional approaches in social theory and in the context of historical collisions of the early twentieth century, had a positive potential, partially realized in the policy of the Soviet state. But the conceptual limitations imposed by the inadequate theoretical basis — the theory of civilizations existing at that time — did not allow the formation of a single Eurasian cultural space to be completed. The Eurasian unity represented by the Soviet Union was consolidated by institutional means, but it was not realized as a cultural synthesis. Chinese scientists have proposed an alternative project for the integration of the Eurasian space — the “One Belt — One Road”, which began to be implemented already in the XXI century. This project is based on Marxist theory and concepts of the second half of the twentieth century, complimentary to Marxism or being neo-Marxist.Conclusions. The incompleteness and instability of the Eurasian project in the Soviet version is a consequence of the limitations inherent in the geopolitical theory itself, which does not take into account the positive potential of Marxist theory and social concepts developed in the second half of the twentieth century. Chinese scientists use the scientific potential of Eurasianism in interpreting the Russian mentality and consider the politics of modern Russia through the prism of Eurasian connotations, but Eurasianism as a geopolitical theory, in their opinion, has not passed the test of history. Therefore, Chinese policy in the Eurasian space is based on other theoretical foundations — in addition to classical Marxism, neo-Marxist approaches. The Chinese authors conclude, in this regard, that Russia is still facing the problem of choosing a geopolitical strategy, which in turn is due to the paradigmatic uncertainty of Russian science.

Within the project “Pragmatics above grammar: Subjectivity and intersubjectivity in Estonian registers and text types” (PRG341) we are studying the expression of subjectivity and intersubjectivity in different written and spoken registers of modern Estonian. We focus on adverbs that function as discourse markers (e.g. vist ‘maybe, probably’, ilmselt ‘apparently, obviously’, tegelikult ‘actually’), markers that develop from main clauses containing cognition verbs that take sentence complements (e.g. (ma) arvan ‘I think’, usun ‘I believe’, (mulle) tundub ‘it seems (to me), it appears (that)’) as well as modal and performative verbs (e.g. võib (juhtuda) ‘can (happen)’, peaks (tulema) ‘should (come)’; kinnitan/väidan (olevat) ‘I affirm/claim’). The analysis combines quantitative corpus-linguistic and qualitative pragmatic approaches, thus belonging to the field of corpus pragmatics. Unlike previous studies of related topics, the project systematically compares the usage of markers in different registers (spoken, online communication, print texts) and text types. The pilot studies performed thus far have revealed several problems with the existing Estonian corpora, important in the study of pragmatics. Firstly, some text types are underrepresented or not represented at all, the text types cannot always be distinguished, and the particular text may not always correspond to the nominal text type (e.g. an academic text may contain quotes from texts of other types). All of this makes it difficult to do comparative statistical analysis of different text types. Secondly, the markers under examination are multifunctional and identifying their (inter)subjective function requires consideration of context broader than a single sentence. However, the public search systems for the existing corpora do not provide this context. For instance, the discourse marker function of cognition verbs is indicated primarily by the fact that the topic of the conversation or text follows through the subordinate clause, not the main clause. Since the available search systems do not provide context larger than a single sentence, the identification of the topic of the discourse, and therefore of the potential discourse-marker function of the verb, is made more difficult. To avoid these problems, the project working group is developing a new “Pragmatics” corpus, being created in the SketchEngine environment. The corpus is made up of 10 subcorpora representing different text types and registers. Each subcorpus contains roughly 500,000 words.

A novel CD34-derived hinge for rapid and efficient detection and enrichment of CAR T cells

Immunotherapy including chimeric antigen receptor (CAR) T cell therapy has revolutionized modern cancer therapy and has achieved remarkable remission and survival rates for several malignancies with historically dismal outcomes. The hinge of the CAR connects the antigen binding to the transmembrane domain and can be exploited to confer features to CAR T cells including additional stimulation, targeted elimination or detection and enrichment of the genetically modified cells. For establishing a novel hinge derived from human CD34, we systematically tested CD34 fragments of different lengths, all containing the binding site of the QBend-10 monoclonal antibody, in a FMC63-based CD19 CAR lentiviral construct. A final construct of 99 amino acids called C6 proved to be the best candidate for flow cytometry-based detection of CAR T cells and >95% enrichment of genetically modified T cells on MACS columns. The C6 hinge was functionally indistinguishable from the commonly used CD8α hinge in vitro as well as in in vivo experiments in NSG mice. We also showed that the C6 hinge can be used for a variety of different CARs and mediates high killing efficacy without unspecific activation by target antigen-negative cells, thus making C6 ideally suited as a universal hinge for CARs for clinical applications.

Organisational Dynamics Founded on the Cross-Functional Development

This paper presents the issues encountered in the adoption of the crossfunctional organization type by the modern enterprises, since the old bureaucratic organization is not any more suitable to the present environment complexity. Apparently, the cross-functional organization seems to be a utopia, due to its difficult implementation. The cross-functional organization is possible only when an enterprise is able to develop cross-functional states of action: decomposition and recomposition of processual organization, of enterprise assembly and of structural organization. An achievable solution for the cross-functional organization of the enterprise is possible if the cohabitation between the old and new organization types of is preserved. The system of cross-functional organization may include the subsystems: operational, information and communication, and instrumental. The cross-functional organization also requires a careful investigation of its internal and external environment.

Unmasking an Enigma: Who Was Lady Wallace and What Did She Achieve?

Lady Wallace had a remarkable life but she has remained a tantalizingly enigmatic figure, not least because we have nothing written in her own hand. She was born in Paris to unmarried parents, a factotum and a linen maid, in 1819. She lived in England from 1872 until her death in 1897, but had a reputation here for speaking only French. Yet she bequeathed to the British nation an art collection described by Lord Rosebery in 1900 as ‘the greatest gift, I believe, that has ever been made by an individual to our country’: the Wallace Collection. Assessing her motivation in making the bequest is not straightforward. While it has been generally assumed that she was simply fulfilling her late husband Sir Richard’s wishes, information circulating at the time and published by the Wallace Collection for many years afterwards stated that she made the bequest at the suggestion of the Wallaces’ private secretary and her residuary legatee, Sir John Murray Scott. Another dichotomy concerns her personality and taste: was she intellectually unremarkable and lacking in discernment, as Baron Ferdinand de Rothschild judged her, or ‘a very refined, shy and excellent lady’, as described by Lord Esher? This article will assess Lady Wallace and her achievements within the context of her time, before considering her afterlife at the Wallace Collection and the role of her bequest in influencing later women patrons of the arts.
