In previous researches, the results showed that selenium Hericium erinaceus polysaccharide and Hericium erinaceus polysaccharide-loaded poly (lactic-co-glycolic acid) nanoparticles enhanced immune responses. In order to further enhance the immune adjuvant activity and phagocytosis of the nanoparticles, two way of combination (selenium-HEP loaded PLGA nanoparticles and selenium modified HEP-PLGA nanoparticles) were prepared to investigate the effects on macrophages in vitro. After treatment with the nanoparticles, the effects of phagocytosis, co-stimulatory molecules expression, nitric oxide (NO), and cytokines secretion were evaluated. The results showed that the particle size, PDI and zeta potential of the selenium-HEP loaded PLGA nanoparticles (Se-HEP-PLGA) and selenium modifified HEP-PLGA nanoparticles (HEP-PLGA-Se) were presented. Se-HEP-PLGA and HEP-PLGA-Se nanoparticles significantly stimulated phagocytic activity, CD40 and CD86 expression of macrophages. In addition, the levels of NO, TNF-α, IL-1β and IL-6 were enhanced in the peritoneal macrophages by stimulation with Se-HEP-PLGA and HEP-PLGA-Se nanoparticles. Among them, Se-HEP-PLGA showed the best effects on the expression of co-stimulatory molecules, secretions of NO and cytokines. These results indicated that Se-HEP-PLGA could enhance the activation of macrophages, and it could be potentially used as an HEP delivery system for the induction of strong immune responses.

Chemoresistance is a major obstacle in chemotherapy of laryngeal carcinoma. Recently, studies indicate that cancer stem cells are responsible for chemotherapy failure. In addition, microRNAs play important roles in tumor initiation, development and multidrug resistance. In the present study, we found that the expression of microRNA-125a was decreased in laryngeal carcinoma tissues and Hep-2 laryngeal cancer stem cells (Hep-2-CSCs). MicroRNA-125a gain-of-function significantly increased the sensitivity of Hep-2-CSCs to cisplatin in vitro and in vivo. Combination with microRNA-125a mimics can decrease the half maximal inhibitory concentration of Hep-2-CSCs to cisplatin. Mechanically, we found that microRNA-125a reverses cisplatin resistance in Hep-2-CSCs by targeting Hematopoietic cell-specific protein 1-associated protein X-1 (HAX-1). Inhibition of HAX-1 by microRNA-125a significantly promotes the cisplatin-induced apoptosis in Hep-2-CSCs through mitochondrial pathway. In addition, multidrug resistance of Hep-2-CSCs to vincristine, etoposide and doxorubicin was greatly improved after the cells were transfected with microRNA-125a mimics. These dates strongly suggested the promotion of microRNA-125a/HAX-1 axis on chemotherapy of laryngeal carcinoma.

The relationship between ongoing brain interoceptive signals and emotional processes has been addressed only indirectly through external stimulus-locked measures. In this study, an internal body trigger (heart evoked potential, HEP) was used to measure ongoing internally triggered signals during emotional states. We employed high-density electroencephalography (hd-EEG), source reconstruction analysis, and behavioral measures to assess healthy participants watching emotion-inducing video-clips (positive, negative, and neutral emotions). Results showed emotional modulation of the HEP at specific source-space nodes of the fronto-insulo-temporal networks related to affective-cognitive integration. This study is the first to assess the direct convergence among continuous triggers of viscerosensory cortical markers and emotion through dynamic stimuli presentation.
