Inhibiting MiR-33a-3p Expression Fails to Enhance ApoAI-Mediated Cholesterol Efflux in Pro-Inflammatory Endothelial Cells

Background and Objectives: Atherosclerosis is an inflammatory condition that results in cholesterol accumulating within vessel wall cells. Atherosclerotic cardiovascular disease is the leading cause of mortality worldwide due to this disease being a major contributor to myocardial infarctions and cerebrovascular accidents. Research suggests that cholesterol accumulation occurring precisely within arterial endothelial cells triggers atherogenesis and exacerbates atherosclerosis. Furthermore, inflamed endothelium acts as a catalyst for atherosclerotic development. Therefore, enhancing cholesterol removal specifically in pro-inflammatory endothelial cells may be a potential treatment option for atherosclerosis. While we have previously shown that inhibiting the microRNA guide strand miR-33a-5p within pro-inflammatory endothelial cells increases both ABCA1 expression and apoAI-mediated cholesterol efflux, it is unknown whether inhibiting the miR-33a-3p passenger strand in pro-inflammatory endothelial cells causes similar atheroprotective effects. In this study, this is what we aimed to test. Materials and Methods: We used plasmid transfection to knockdown miR-33a-3p expression within cultured pro-inflammatory immortalized mouse aortic endothelial cells (iMAECs). We compared ABCA1 expression and apoAI-mediated cholesterol efflux within these cells to cultured pro-inflammatory iMAECs transfected with a control plasmid. Results: The knockdown of miR-33a-3p expression within pro-inflammatory iMAECs resulted in a significant increase in ABCA1 mRNA expression. However, the inhibition of miR-33a-3p did not significantly increase ABCA1 protein expression within pro-inflammatory iMAECs. Moreover, we failed to detect a significant increase in apoAI-mediated cholesterol efflux within pro-inflammatory iMAECs from miR-33a-3p knockdown. Conclusions: Our results indicative that the knockdown of miR-33a-3p alone does not enhance ABCA1-dependent cholesterol efflux within pro-inflammatory endothelial cells. To gain any atheroprotective benefit from inhibiting miR-33a-3p within pro-inflammatory endothelium, additional anti-atherogenic strategies would likely be needed in unison.

The Short-Term Effects of Altering Milking Intervals on Milk Production and Behavior of Holsteins Milked in an Automated Milking System

The widespread adoption of automatic milking systems (AMS) in the United States has afforded dairy cows the flexibility to establish personalized milking, feeding, and resting schedules. Our study focused on investigating the short-term effects of transitioning milking permissions from every 4 (MP4) to 6 (MP6) hours on the 100th day of lactation on milking frequency, milk yields, and cow behavior. Twenty-four Holstein dairy cows were divided into control (maintaining a 4 h milking interval) and test groups (transitioning to a 6 h milking interval) and observed for 6 days. The analysis revealed that parity and treatment had no significant impact on milking frequency, milk/visit, or daily milk yield. However, multiparous cows spent more time inside the commitment pen, while test group cows exhibited more tail-swishing and displacement behavior, approached the AMS more frequently, and spent longer idle times. The interaction between parity and treatment influenced heart rate variability parameters, indicating increased stress in the test group cows. Additionally, the test group cows had greater total and daytime lying frequencies, suggesting short-term behavioral modifications. Despite no immediate impact on milk production, further research is recommended to assess the potential long-term effects on milk yield in AMS farms, considering the identified stress indicators short-term.

Sostenibilidad empresarial aplicada al Diseño Gráfico

El artículo presenta la llegada del nuevo milenio, un número cada vez mayor de empresarios se unieron a la aplicación del diseño sostenible que comenzó a replantearse en las empresas y el rol que juegan con el desarrollo del medio ambiente, el planeta y en la sociedad. Podemos decir que el diseño sostenible busca generar soluciones a través de servicios y estilos de vida, pero no exclusivamente a través de objetos. Con el fin de introducir una definición elaborada de diseño sostenible es necesario mencionar los sistemas sostenibles, que básicamente, se refieren a cualquier tipo de red o servicio social que puede existir y replicarse. Además de sistemas sostenibles hay otros principios dentro del diseño sostenible. Por último, cualquier tipo de resultado obtenido para satisfacer la necesidad debe ser sostenible a largo plazo entendiéndose como un proceso que permita una comunidad lograr un resultado a través de estrategias de diseño.
