Effect of breed and dietary composition on the miRNA profile of beef steers divergent for feed efficiency

Abstract Identifying and breeding cattle that are more feed efficient is of great benefit to beef production. Additionally, it is crucial that genes contributing to feed efficiency are robust across varying management settings including dietary source as well as being relevant across contrasting breeds of cattle. The aim of this study was to determine miRNAs that are contributing to the expression of residual feed intake (RFI) across two breeds and dietary sources. miRNA profiling was undertaken in Longissimus dorsi tissue of Charolais and Holstein–Friesian steers divergent for RFI phenotype following two contrasting consecutive diets (high-forage and high-concentrate). Ten miRNA were identified as differentially expressed (adj. P < 0.1) across the breed and diet contrasts examined. Of particular interest was the differential expression of miR-2419-5p and miR-2415-3p, both of which were up-regulated in the Low-RFI Charolais steers across each dietary phase. Pathway analysis of target mRNA genes of differentially expressed miRNA revealed enrichment (P < 0.05) for pathways including metabolic related pathways, insulin receptor signalling, adipogenesis as well as pathways related to skeletal muscle growth. These results provide insight into the skeletal muscle miRNAome of beef cattle and their potential molecular regulatory mechanisms relating to feed efficiency in beef cattle.

A One Health framework for exploring zoonotic interactions demonstrated through a case study

Abstract The eco-epidemiology of zoonoses is often oversimplified to host-pathogen interactions while findings derived from global datasets are rarely directly transferable to smaller-scale contexts. Through a systematic literature search, we compiled a dataset of naturally occurring zoonotic interactions in Austria, spanning 1975–2022. We introduce the concept of zoonotic web to describe the complex relationships between zoonotic agents, their hosts, vectors, food, and environmental sources. The zoonotic web was explored through network analysis. After controlling for research effort, we demonstrate that, within the projected unipartite source-source network of zoonotic agent sharing, the most influential zoonotic sources are human, cattle, chicken, and some meat products. Analysis of the One Health 3-cliques (triangular sets of nodes representing human, animal, and environment) confirms the increased probability of zoonotic spillover at human-cattle and human-food interfaces. We characterise six communities of zoonotic agent sharing, which assembly patterns are likely driven by highly connected infectious agents in the zoonotic web, proximity to human, and anthropogenic activities. Additionally, we report a frequency of emerging zoonotic diseases in Austria of one every six years. Here, we present a flexible network-based approach that offers insights into zoonotic transmission chains, facilitating the development of locally-relevant One Health strategies against zoonoses.

Assessment of in vitro digestibility and postdigestion peptide release of mare milk in relation to different management systems and lactation stages

ABSTRACT: Mare milk has a unique protein composition that makes it a preferred option for adult and infant nutrition. Several functional properties have been attributed to this milk, but with little current evidence. In fact, knowledge on mare milk composition is still limited. In particular, studies addressing the performance of mare milk proteins during human gastrointestinal digestion are scarce, which limits the understanding of mare milk nutritional quality and functionality. For this reason, the present study describes the digestibility of mare milk proteins and the release of peptides as affected by management and lactation stage, factors which are known to affect milk composition. Mare milk samples from 3 different farms collected during 6 mo of lactation (n = 54) were subjected to a static in vitro gastrointestinal model to measure peptide release and protein digestibility. In the present study, a detailed description of protein and individual amino acid behavior during the digestion process was given. For the first time, digestion of the 2 equine β-lactoglobulin isoforms (I and II) was described individually. In addition, it was found that lactation stage and management system can significantly affect protein digestibility and peptide release during gastrointestinal digestion of mare milk. Presumably, differences in the composition of mare milk influence the protein structure and enzyme accessibility, which might have an impact on digestion behavior. Although no specific bioactive peptides were identified, several precursors of previously described bioactive peptides were found. These findings could support the idea of mare milk as a food with added value.

Heritabilities and genetic and phenotypic correlations for milk production and fertility traits of spring-calved once-daily or twice-daily milking cows in New Zealand

ABSTRACT: The objectives of this study were to estimate the genetic and phenotypic correlations and heritabilities for milk production and fertility traits in spring-calved once-daily (OAD) milking cows for the whole season in New Zealand and compare those estimates with twice-daily (TAD) milking cows. Data used in the study consisted of 69,252 first parity cows from the calving seasons 2015–2016 to 2017–2018 in 113 OAD and 531 TAD milking herds. Heritability estimates for production and fertility traits were obtained through single-trait animal models, and estimates of genetic and phenotypic correlations were obtained through bivariate animal models. Heritability estimates of production traits varied from 0.26 to 0.61 in OAD and from 0.13 to 0.63 in TAD. Heritability estimates for fertility traits were low in both OAD and TAD milking cow populations, and estimates were consistent (OAD: 0.01 to 0.10 and TAD: 0.01 to 0.08) across milking regimens. Estimates of phenotypic and genetic correlations among production traits were consistent across populations. In both populations, phenotypic correlations between milk production and fertility traits were close to zero, and most of the genetic correlations were antagonistic. In OAD milking cows, genetic correlations of milk and lactose yields with the start of mating to conception, 6-wk in-calf, not-in-calf, and 6-wk calving rate were close to zero. Interval from first service to conception was negatively genetically correlated with milk and lactose yields in OAD milking cows. Protein percentage was positively genetically correlated with 3-wk and 6-wk submission, 3-wk in-calf, 6-wk in-calf, first service to conception, 3-wk calving, and 6-wk calving rate in the TAD milking cow population, but these correlations were low in the OAD milking cow population. Further studies are needed to understand the relationship of protein percentage and fertility traits in the OAD milking system. The phenotypic correlations between fertility traits were similar in OAD and TAD milking populations. Genetic correlations between fertility traits were strong (≥0.70) in cows milked TAD, but genetic correlations varied from weak to strong in cows milked OAD. Further research is required to evaluate the interaction between genotype by milking regimen for fertility traits in terms of sire selection in the OAD milking cow population.

Development of a novel stall design for dairy cattle: Part II. The effect of minimal stall partitioning on lying behavior, rumination, stall cleanliness, and preference

Free-stalls for dairy cows promote cleanliness, provide cows with a defined space to lie down, and decrease labor and bedding required for maintenance. However, current stall features can restrict behavior and reduce stall use. The objective of this study was to assess the short-term effect of a novel free-stall design (flexible single-bar partition, no neck rail, increased slope) on stall cleanliness and the lying behavior, rumination, milk production, and preferences of dairy cows in comparison to standard free-stalls (metal loop partitions, neck rail). In the first experiment, 60 Holstein cows were randomly divided into two groups and switched between standard and novel stalls in a replicated crossover design with four 7-day periods. Lying behavior and rumination were recorded continuously. Milk yields were recorded 2x/day, and stall cleanliness scores were collected on the last four days of weeks 3 and 4. The second experiment was a 6-day preference test where 14 cows from experiment 1 were given free access to 16 standard and 16 novel stalls. Lying behavior was recorded continuously. On day 6, standing, perching, and intention, lying, and rising movements were recorded. In experiment 1, cows spent 12 min less time lying down (14.1 vs 14.3 ± 0.48 h/d), had one fewer lying bouts (8.8 vs 9.8 ± 0.23 bouts/d), and had longer lying bouts by 6 min (1.4 vs 1.5 ± 0.05 h/bout) in novel stalls. Rumination time (547.3 vs 548.9 ± 4.66 min/d) and milk yield (35.0 vs 35.2 ± 0.51 kg/d) did not differ between stall types. However, novel stalls were more than twice as soiled as standard stalls (32.8 vs 14.2 ± 1.38% of rear half soiled). In experiment 2, the cows most recently housed in the novel stalls showed a clear preference for lying down in those stalls, whereas cows most recently housed in standard stalls showed no preference. Intention and lying down movements were longer in standard stalls when other cows were present in diagonally opposite stalls, but were similar between stall types when not occupied. All cows preferred standing in novel stalls. Novel stalls may have improved stall use compared to standard free-stalls, and the design requires further development to address cleanliness concerns. Future work is required to provide comfortable stalls without sacrificing cleanliness.

Monomorphic allele rs109231213 in 3’UTR PLAG1 gene in purebred of Bali cattle (Bos javanicus)

The mutation rs109231213 that is located in 3’UTR of PLAG1 gene is associated with the growth and body weight in several Bos taurus and Bos indicus breeds. This study aimed to identify SNP rs109231213 in Bali cattle (Bos javanicus). The study used 41 samples of Bali cattle. The PLAG1 gene polymorphism was analyzed using PCR and direct sequencing methods. PCR pimers were 5’- TTGCACAGAATCAGTGTGTC-3’ and 5’- AGCCTAACGTGGATCTATGG-3’. The results showed that primers successfully amplified the 331 bp fragment at annealing 60°C that contained rs109231213. SNP was monomorphic in Bali cattle with one allele (G). This study concludes that rs109231213 in 3’UTR of PLAG1 gene can be used as specific marker in purebred of Bali cattle that have never been crossed with Bos taurus and Bos indicus.

Mutational and phylogenetic status of west siberian strains of BLV

The study is devoted of full-genome BLV sequences circulating in cattle populations of the Novosibirsk region, Russia. The phylogenetic tree shows that the West Siberian isolates are quite closely related to such previously isolated strains as AF399704 (Brazil), AP018007, AP018016, AP018019, LC007988, LC007991 (Japan) and EF065638 (Belgium) we calculations show that the number of mutations that could independently occur in parallel evolving BLV strains significantly exceeds the expected number based on the probability of corresponding substitutions. It was also found that the studied isolates have some mutations, the presence of which, at first glance, is possible only with their divergent development in different independently evolving branches. However, calculations show that the probability of an independent origin of an identical mutation is extremely small, which indicates the possibility of exchanging RNA sites between isolates circulating in West Siberian cattle populations.

Understanding breeding preferences among small-scale cattle producers: implications for livestock improvement programmes

Livestock production provides a pathway for improving livelihoods and reducing poverty in semi-arid tropical regions. However, this contribution has been affected by low livestock productivity. Most livestock programmes have also failed due to, among other things, the inability to understand the dynamics in smallholder breeding preferences. Using data from the sub-humid region in Zimbabwe, this paper sought to provide evidence on smallholder cattle breeding preferences and the implication on livestock improvement programmes. It applies the choice experiment approach to model farmer preferences for selected cattle breeding attributes. The results show three attributes that significantly affect breeding preferences. The attributes ‘cow body condition score’ and the ‘useful life of a bull/semen’ have a positive influence while ‘artificial insemination/bull maintenance cost’ negatively affects farmer preferences. This means farmers prefer breeding strategies which improve the nutrition of their cows, have a longer lifespan for the bull/semen and whose cost of breeding services is low. However, access to education and income affected these preferences. Education made farmers to make informed choices while higher incomes increased the propensity of investing in livestock breeding technologies. The findings also show that existing institutional arrangements in animal management and community grazing do not promote investment in livestock improvement. Thus, more attention should be given to improving animal nutritional management which includes promoting sustainable grazing schemes. There is also a need to provide affordable livestock breeding services through recruiting and training more artificial insemination service providers. Strong and effective institutions that provide incentives for collective participation are integral to any community-based livestock breeding programme. There is also a need to promote access to information and enhance farmers’ knowledge and capacity in improved livestock management practices.

Choices between red clover and fescue in the diet can be reliably estimated in heifers post-weaning using n-alkanes

Measuring diet choice in grazing animals is challenging, complicating the assessment of feed efficiency in pasture-based systems. Furthermore, animals may modify their intake of a forage species depending on its nutritive value and on their own physiological status. Various fecal markers have been used to estimate feed intake in grazing animals. However, plant-wax markers such as n-alkanes (ALK) and long-chain alcohols may provide reliable estimates of both dietary choices and intakes. Still, their use in beef cattle has been relatively limited. The present study was designed to test the reliability of the ALK technique to estimate diet choices in beef heifers. Twenty-two Angus-cross heifers were evaluated at both post-weaning and yearling age. At each age, they were offered both red clover and fescue hay as cubes. Following 3-week acclimation periods, daily intake of each forage species was assessed daily for 10 days. During the final 5 days, fecal grab samples were collected twice daily. The ALK fecal concentrations were adjusted using recovery fractions compiled from literature. Diet composition was estimated using two statistical methods. Post-weaning, dietary choices were reliably estimated, with low residual error, regardless of the statistical approach adopted. The regression of observed on estimated red clover proportion ranged from 0.85±0.08 to 1.01±0.09 for fecal samples collected in the p.m. and for daily proportions once averaged, respectively. However, at yearling age, the estimates were less reliable. There was a tendency to overestimate the red clover proportion in diets of heifers preferring fescue, and vice versa. This was due to greater variability in ALK fecal concentrations in the yearling heifers. Overall, the ALK technique provided a reliable tool for estimating diet choice in animals fed a simple forage diet. Although further refinements in the application of this methodology are needed, plant-wax markers provide opportunities for evaluating diet composition in grazing systems in cattle.

Methods and consequences of including reduction in greenhouse gas emission in beef cattle multiple-trait selection

Abstract Background Societal pressures exist to reduce greenhouse gas (GHG) emissions from farm animals, especially in beef cattle. Both total GHG and GHG emissions per unit of product decrease as productivity increases. Limitations of previous studies on GHG emissions are that they generally describe feed intake inadequately, assess the consequences of selection on particular traits only, or examine consequences for only part of the production chain. Here, we examine GHG emissions for the whole production chain, with the estimated cost of carbon included as an extra cost on traits in the breeding objective of the production system. Methods We examined an example beef production system where economic merit was measured from weaning to slaughter. The estimated cost of the carbon dioxide equivalent (CO2-e) associated with feed intake change is included in the economic values calculated for the breeding objective traits and comes in addition to the cost of the feed associated with trait change. GHG emission effects on the production system are accumulated over the breeding objective traits, and the reduction in GHG emissions is evaluated, for different carbon prices, both for the individual animal and the production system. Results Multiple-trait selection in beef cattle can reduce total GHG and GHG emissions per unit of product while increasing economic performance if the cost of feed in the breeding objective is high. When carbon price was $10, $20, $30 and $40/ton CO2-e, selection decreased total GHG emissions by 1.1, 1.6, 2.1 and 2.6% per generation, respectively. When the cost of feed for the breeding objective was low, selection reduced total GHG emissions only if carbon price was high (~ $80/ton CO2-e). Ignoring the costs of GHG emissions when feed cost was low substantially increased emissions (e.g. 4.4% per generation or ~ 8.8% in 10 years). Conclusions The ability to reduce GHG emissions in beef cattle depends on the cost of feed in the breeding objective of the production system. Multiple-trait selection will reduce emissions, while improving economic performance, if the cost of feed in the breeding objective is high. If it is low, greater growth will be favoured, leading to an increase in GHG emissions that may be undesirable.

<b>Appropriate mathematical models to describe the lactation curves of milk production traits of Iranian Holstein dairy cattle

The objective of this study was to compare the goodness of fit of lactation curve models; Wood, Wilmink, Linear Splines (SPL), Cubic Splines (SPC), Quadratic Splines (SPQ), Cobby and Le Du, Ali Schaeffer and Legendre Polynomial (LEG), in random regression model (RRM) for milk production traits of Iranian Holstein dairy cattle. For this purpose the records obtained from Test-day (TD) regarding milk (928513), fat (788577) and protein (653317) yields related to their first parity were used. These data collected from the years of 2003 to 2011 by the Karaj breeding center of Iran. The genetic parameters were estimated using REML method using WOMBAT software. Based on obtained results, RRM with SPL6 (6,6), SPC6 (6,6) and LEG (3,5) for milk yield, SPL6 (6,6), SPQ6 (6,6), LEG (3,5) for fat yield and SPL5 (5,5), SPQ4 (4,4) and LEG (3,4) for protein yield, were selected as better model to describe the lactation curves. The estimated heritabilities by best models were lower in the beginning lactation than other during lactation. The genetic trend of milk yields was showed an increasing during the 10 past years, which indicated Iranian Holstein dairy cattle population genetically was improved for milk yields.

A novel immunotherapy of Brucellosis in cows monitored non invasively through a specific biomarker.

Brucellosis is an important zoonotic disease causing huge economic losses worldwide. Currently no effective immunotherapy for Brucellosis or any biomarker to monitor the efficacy of therapy is available. Treatment is ineffective and animals remain carrier lifelong. S19 and RB51 are live attenuated vaccine strains of Brucella abortus. However, S19 induces only antibody, ineffective for intracellular pathogen. RB51 induces cell mediated immunity (CMI) but it is Rifampicin resistant. Both organisms are secreted in milk and can infect humans and cause abortions in animals. Phage lysed bacteria (lysates) retain maximum immunogenicity as opposed to killing by heat or chemicals. We report here the successful immunotherapy of bovine Brucellosis by phage lysates of RB51 (RL) and S19 (SL). The SL induced strong antibody response and RL stimulated CMI. In vitro restimulation of leukocytes from RL immunized cattle induced interferon gamma production. A single subcutaneous dose of 2 ml of cocktail lysate (both RL and SL), eliminated live virulent Brucella from Brucellosis affected cattle with plasma level of Brucella specific 223 bp amplicon undetectable by RT-PCR and blood negative for live Brucella by culture in 3 months post-immunization. This is the first report on minimally invasive monitoring of the efficacy of antibacterial therapy employing plasma RNA specific for live bacteria as a biomarker as well as on the use of RB51 phage lysate for successful immunotherapy of Brucellosis in cattle.

Current situation and future prospects for beef production in China — A review

The beef industry is an important part of livestock and meat production in China. China ranks third in the world for beef production. With the rapid development of the Chinese economy, beef consumption has grown rapidly, and beef consumption has been increasing with rising per capita gross domestic production. However, the domestic beef industry in China has not been able to keep pace with growth in consumption, making China a net importer of beef from other countries. Moreover, the volume of production has increased little despite rising demand. The slowing of growth in beef production in recent years has led to a sharp rise in beef prices. Domestic beef production and consumption is restricted by a shortage of beef cattle inventory. The Chinese beef industry is facing many technical problems including transformation of traditional practices, feeding and management systems, and genetic improvement of cattle breeds. The long-term, sustainable development of the Chinese beef industry is an important issue for China.

Comparative study on production, reproduction and functional traits between Fleckvieh and Braunvieh cattle

Objective Aim of the current comparative study was to evaluate production outputs, reproduction efficiency and functional traits in dual-purpose Fleckvieh and Braunvieh cows, reared under temperate European conditions. Methods A data-set from 414 Fleckvieh and 42 Braunvieh cows and 799 lactations was analysed. ID tag number, milk yield per milking session, number of steps/interval and milk conductivity were recorded and collected daily using AfiMilk 3.076 A-DU software (Afimilk Ltd., Kibbutz, Israel). Production and milk quality data were taken from the results of the official performance recordings and the reproductive outputs of cows were recorded by the research stations veterinarians. Comparisons between the two genotypes were carried out using the one way analysis of variance protocol, with categorical factor being considered the breed of cows. All the statistical inferences were carried out using Statistica software (StatSoft Inc., Tulsa, OK, USA). Results Fleckvieh cows significantly outperformed (p≤0.05) the Braunvieh herd, with average milk yields of 5,252.1±35.79 kg and 4,897.6±128.94 kg, respectively. Age at first calving was significantly (p≤0.01) influenced by the breed, with Fleckvieh heifers being more precocious (32.8±0.29 mo) compared to those of Braunvieh breed (35.7±0.84 mo). Reproduction efficiency as defined by the number of inseminations per gestation, calving interval, dystocia, days dry and days open, was not influenced by genotype (p>0.05). Incidences of sub-clinical mastitis, clinical mastitis, lameness and abortions were not influenced by the breed factor (p>0.05). Stayability of cows was significantly (p≤0.001) influenced by genotype, with Braunvieh cows having an average age at culling of 117.88±11.78 months compared to 90.88±2.89 months in Fleckvieh. Conclusion Overall, results have shown that genotype significantly influenced milk yield, age at first calving and longevity.

Interaction of bovine peripheral blood polymorphonuclear cells and Leptospira species; innate responses in the natural bovine reservoir host.

Cattle are the reservoir hosts of Leptospira borgpetersenii serovar Hardjo, and can also be reservoir hosts of other Leptospira species such as L. kirschneri, and L. interrogans. As a reservoir host, cattle shed Leptospira, infecting other animals, including humans. Previous studies with human and murine neutrophils have shown activation of neutrophil extracellular trap or NET formation, and upregulation of inflammatory mediators by neutrophils in the presence of Leptospira. Humans, companion animals and most widely studied models of Leptospirosis are of acute infection, hallmarked by systemic inflammatory response, neutrophilia and septicemia. In contrast, cattle exhibit chronic infection with few outward clinical signs aside from reproductive failure. Taking into consideration that there is host species variation in innate immunity, especially in pathogen recognition and response, the interaction of bovine peripheral blood polymorphonuclear cells (PMNs) and several Leptospira strains was evaluated. Studies including bovine-adapted strains, human pathogen strains, a saprophyte and inactivated organisms. Incubation of PMNs with Leptospira did induce slight activation of neutrophil NETs, greater than unstimulated cells but less than the quantity from E. coli P4 stimulated PMNs. Very low but significant from non-stimulated, levels of reactive oxygen peroxides were produced in the presence of all Leptospira strains and E. coli P4. Similarly, significant levels of reactive nitrogen intermediaries (NO2) was produced from PMNs when incubated with the Leptospira strains and greater quantities in the presence of E. coli P4. PMNs incubated with Leptospira induced RNA transcripts of IL-1β, MIP-1α, and TNF-α, with greater amounts induced by live organisms when compared to heat-inactivated leptospires. Transcript for inflammatory cytokine IL-8 was also induced, at similar levels regardless of Leptospira strain or viability. However, incubation of Leptospira strains with bovine PMNs did not affect Leptospira viability as measured by limiting dilution culture. This is in contrast to previously reported results of innate inflammatory activation by Leptospira in human and other animal models, or the activation and interaction of bovine PMNs with Escherichia coli and other bacterial pathogens.
